It has been found that a variety of stimuli including danger-associated molecular patterns DAMPs, such as silica and uric acid crystals and pathogen-associated molecular patterns PAMPs can activate NLRP3 inflammasome, but the specific regulatory mechanisms of NLRP3 inflammasome activation remain unclear. In this review, we summarize current understanding of the regulatory mechanisms of NLRP3 inflammasome activation as well as inhibitors that specifically and directly target NLRP3. The NLRP3 inflammasome can trigger inflammation by sensing a wide range of stimuli, but the specific mechanisms are still unclear.

Cryopyrin is found mainly in white blood cells and in cartilage-forming cells chondrocytes. NLR proteins are involved in the immune system, helping to start and regulate the immune system's response to injury, toxins, or foreign invaders. NLR proteins recognize specific molecules and respond by helping to turn on activate certain parts of the immune system. Cryopyrin recognizes bacteria; chemicals such as asbestos, silica, and uric acid crystals; and compounds released by injured cells. Cryopyrin molecules assemble themselves, along with other proteins, into structures called inflammasomes, which help trigger the process of inflammation. Inflammation occurs when the immune system sends signaling molecules as well as white blood cells to a site of injury or disease to fight foreign invaders and help repair damaged tissues. Once the threat is over, the body stops inhibits the inflammatory response, to prevent damage to its own cells and tissues. CAPS are a group of conditions that have overlapping signs and symptoms. The conditions are generally characterized by periodic episodes of skin rash, fever, and joint pain. These conditions were once thought to be distinct disorders but are now considered to be part of the same condition spectrum. All of the variants likely result in the cryopyrin protein being overactive.

NLRP3 NOD-, LRR- and pyrin domain-containing protein 3 is an intracellular sensor that detects a broad range of microbial motifs, endogenous danger signals and environmental irritants, resulting in the formation and activation of the NLRP3 inflammasome.

In this review, we discuss the current understanding of the mechanistic details of NLRP3 inflammasome activation with a particular emphasis on protein-protein interactions, posttranslational modifications, and spatiotemporal regulation of the NLRP3 inflammasome machinery.

Disruption of Nrf2, a key inducer of antioxidant defenses, attenuates ApoE-mediated atherosclerosis in mice. Schmid-Burgk J. Menu P. Mridha, A. Duong, B. Fernandes-Alnemri T. Bacterial exotoxins and the inflammasome. Early epithelial necrosis was more severe in infected mutant mice, with extensive collagen deposition leading to later respiratory compromise, suggesting a function for the cryopyrin inflammasome in healing responses. Among these inflammasomes, the NLRP3 inflammasome is the most extensively studied. Clapham D.
